Propuesta de medio de cultivo para el estudio de Phytophthora Nicotianae Breda de Haan

Abstract

El género Phytophthora representa un grupo difícil de aislar y conservar. Por lo que se han desarrollado varios medios de cultivo para su estudio. Se propone al medio Agar-tabaco como alternativa para el crecimiento y desarrollo de P. nicotianae. Para realizar este trabajo se emplearon dos cepas: Pp1 y Pp6 aislados de tabaco y piña respectivamente. Ambas se sembraron en discos en agar-tabaco, y se comparó con agar tomate y agar harina de maíz. Se determinó el crecimiento lineal del hongo, las características del micelio y estructuras formadas en cada medio. El crecimiento micelial fue graficado y analizado estadísticamente, los datos restantes fueron tabulados. Pp1 creció aceleradamente en el medio agar tabaco mostrando diferencias significativas con el resto. Pp6 tuvo un crecimiento lento en agar tabaco con diferencias significativas. Ambas cepas mostraron un micelio no algodonoso, pobre y con bordes irregulares; formaron estructuras reproductivas como clamidosporas y esporangios. Se considera el medio agar tabaco alternativo para el crecimiento de P. nicotianae aislado de diferentes hospederos.

Phytophthora genus has been a group whose isolation and conservation is laborious. For this reason, several culture medium have been developed for their study. This paper proposes the tobacco-agar medium as alternative for the growth and development of P. nicotianae. In this work the strains Pp1 and Pp 6 isolated of tobacco and pineapple respectively, were used. The isolates were placed individually in Petri dishes with tobacco-agar medium, tomato-agar flour medium and cornmeal-agar medium. The cultural and morphological characteristics were described and the lineal growth of the fungus was measured and graphed. Pp1 grew rapidly in tobacco-agar medium, showing significant differences with the rest of the media tried. Pp6 showed a significantly minor growth in tobacco-agar medium. Both strains showed a weak, fluffy micelia with irregular borders; they formed reproductive structures as chlamydospore and sporangia. The tobacco-agar medium was considered effective for growth of P. nicotianaeisolated from different hosts.
